Đặc điểm của lan con nuôi cấy mô
Lan con nuôi cấy mô được sinh trưởng trong môi trường nhân tạo, giàu dinh dưỡng, độ ẩm cao, ánh sáng yếu và hoàn toàn vô trùng. Trong điều kiện này, cây phát triển nhanh nhưng hệ rễ và cấu trúc mô còn non yếu, chưa thích nghi với sự thay đổi đột ngột của môi trường bên ngoài. Khi đưa ra trồng thực tế, cây rất dễ bị sốc nhiệt, úng nước, thối rễ hoặc nhiễm nấm bệnh nếu không có giai đoạn chuyển tiếp phù hợp.
Bên cạnh đó, môi trường khí hậu nóng ẩm ở nhiều khu vực, đặc biệt là các đô thị và vùng nhiệt đới, là điều kiện lý tưởng cho vi sinh vật gây bệnh phát triển. Do đó, kỹ thuật trồng lan con từ nuôi cấy mô không chỉ đơn thuần là trồng cây vào chậu mà là một quy trình tổng hợp, bao gồm chuẩn bị vật tư, xử lý cây giống, kiểm soát môi trường và chăm sóc đúng cách trong từng giai đoạn.
Chuẩn bị vật liệu và dụng cụ trồng
Trước khi tiến hành trồng lan con, cần chuẩn bị đầy đủ dụng cụ và vật liệu để đảm bảo quá trình thao tác diễn ra nhanh chóng, chính xác và hạn chế tối đa nguy cơ nhiễm bệnh.
Dụng cụ cần thiết bao gồm móc cấy để lấy lan ra khỏi chai nuôi cấy, dao cắt rễ và kéo cắt cành đã được khử trùng, que nhọn để tạo lỗ trồng, kẹp gắp cây và các loại chậu chung hoặc chậu đơn tùy theo phương án trồng. Tất cả dụng cụ nên được sát trùng bằng cồn hoặc dung dịch chuyên dụng trước khi sử dụng.
Chất trồng đóng vai trò quan trọng trong việc giữ ẩm, thoát nước và tạo điều kiện cho rễ lan phát triển. Các vật liệu thường được sử dụng gồm than gỗ, than bùn, vỏ thông, dớn và cát. Điều kiện môi trường và giai đoạn chuyển tiếp
Lan con dễ chết do úng nước hoặc độ ẩm quá cao kéo dài, đây cũng là nguyên nhân chính gây ra các bệnh thối rữa. Trong phòng thí nghiệm, cây được nuôi ở nhiệt độ ổn định từ 21 đến 23 độ C, pH môi trường khoảng 5,2. Khi đưa ra ngoài, nếu không có bước chuyển tiếp, cây rất dễ bị tổn thương.
Do đó, cần thiết lập một giai đoạn thuần dưỡng trong nhà kính hoặc khu giâm cành. Nhiệt độ trong giai đoạn này không nên vượt quá 25 độ C, độ ẩm không khí duy trì ở mức 80–90%, cường độ ánh sáng khoảng 5.000 lux. Quy trình đưa lan con ra khỏi chai nuôi cấy
Trước tiên, chuẩn bị một chậu nước sạch sử dụng nước máy đã để bay hết clo, điều chỉnh pH khoảng 5,2. Trong nước có thể pha một nồng độ rất loãng thuốc ngừa nấm để phòng bệnh cho cây trong giai đoạn đầu.
Lấy chai nuôi cấy lan con, tháo bông gòn ở cổ chai, cho một ít nước vào và lắc nhẹ để tách cây khỏi môi trường thạch. Sau đó lật ngược chai để cây lan theo nước chảy ra ngoài chậu. Dùng kẹp gắp nhẹ nhàng từng cây, rửa sạch phần thạch còn bám vào rễ bằng cách nhúng qua lại nhiều lần trong nước.
Sau khi làm sạch, tiến hành cắt bỏ toàn bộ rễ hư, rễ dập hoặc có dấu hiệu thối. Đặt cây lên đĩa hoặc khay sạch để ráo nước trước khi trồng. Đây là bước quan trọng giúp hạn chế nấm bệnh phát sinh từ các mô rễ bị tổn thương.
Kỹ thuật trồng lan con vào chậu
Chậu chung hoặc chậu đơn sau khi đã chuẩn bị giá thể, dùng que nhọn tạo lỗ trồng vừa đủ. Đặt cây lan con vào lỗ, nhẹ nhàng dùng tay ấn gốc để giá thể ôm sát bộ rễ nhưng không nén quá chặt. Sau khi trồng xong, rửa nhẹ lại cả chậu và cây để loại bỏ bụi bẩn còn sót.
Tiếp theo, phun cho cây một dung dịch dinh dưỡng loãng, phù hợp với từng loài lan nhằm tăng sức chịu đựng trong giai đoạn đầu. Cần lưu ý hạn chế các dung dịch chứa nhiều chất hữu cơ vì dễ gây thối rễ. Trong trường hợp không có điều kiện, có thể sử dụng phân NPK 30-10-10 với nồng độ rất loãng.

Sau khi trồng, đặt cây ở nơi râm mát, tránh ánh nắng trực tiếp. Nếu trồng trong nhà kính, mỗi ngày chỉ tưới trực tiếp lên cây khoảng 2 lần, những lần còn lại tưới sàn để duy trì độ ẩm không khí. Nếu trồng ở khu vực râm mát ngoài trời, cần tưới tối thiểu 4 lần mỗi ngày, lần tưới cuối cùng nên trước 16 giờ để tránh ẩm độ cao ban đêm.
Nước tưới nên là dung dịch dinh dưỡng đã được điều chỉnh pH phù hợp, không chứa các tạp chất hữu cơ dễ gây bệnh. Sau khoảng một tháng, khi rễ cây phát triển mạnh và bám chắc vào giá thể, có thể chuyển cây ra khỏi nhà kính và bắt đầu chế độ chăm sóc như lan trưởng thành.
